NHP PB MNC are enriched by density separation. NHP BM MNC are isolated from fresh bone marrow by diluting the bone marrow with phosphate-buffered saline and using gradient separation techniques. After centrifugation, the MNC layer is collected. MNC can be processed to isolate subpopulations. NHP BM CD34+ cells are isolated from NHP bone marrow mononuclear cells using a direct immunomagnetic CD34+ MicroBead labeling system. Harvested purified cells are analyzed using a laser analytical flow cytometer confirming the purity is greater than 90%. FACScan will be provided. NHP-Plasma is processed through centrifugation to diminish cells and platelets.

Cryopreservation and percentage used: 50% of Iscove’s DMEM with L-glutamine & 25 mM HEPES, 40% of Fetal Bovine Serum (FBS), 10% of Dimethyl sulfoxide (DMSO).
Commonly used media and buffer in biological research, such like Staining Buffer (SB), Phosphate-Buffered Saline (PBS), Bovine Serum Albumin (BSA), RPMI (without L-glutamine) and Dulbecco’s Modified Eagle’s Medium (DMEM) might be used in the cells isolation processes.
