Alive healthy laboratory mouse obtained from animal laboratories that have not been inoculated with or exposed to any livestock or avian species is conducted or poultry disease agent’s exotic. The fresh spleen is harvested from alive mouse and dissociated from debris. Lysis method is used to eliminate red blood cells and platelets to purify the splenocytes. Washed 4 times after Ficoll-Paque. About 20-30 donor’s splenocytes are pooled for isolation. Natural Killer (NK) cells CD49b+ then are isolated using specific immunomagnetic method with AutoMACS system following the protocol of the manufacturer. Purified of NK cells are analyzed using a laser analytical flow cytometer, confirming the purity to be greater than 80%. FACScan will be provided.

CD49b+ NK Cells were used to study Natural Killer (NK) cell cytotoxicity in vitro or after adoptive transfer of isolated NK cells in vivo. Furthermore, isolated NK cells were used to evaluate their role in experimental mouse models of immune-mediated liver injury and chronic infection.
