Alive healthy laboratory rat obtained from animal laboratories that have not been inoculated with or exposed to any livestock or avian species is conducted or poultry disease agent’s exotic. The fresh cerebellum, cortex, hippocampus is harvested from 24-48 hours’ newborn rat brain. We use enzymatic digestion (trypsin and papain) and mechanical dissociation separation and purification. About 5 donor samples are pooled. Further, the cells are seeded, medium exchange occurred from 3 days after obtaining a culture and after, every other day for about 10 days to reach the confluence. The astrocytes are enriched in vitro, using specific medium and cultural conditions. Harvested purified astrocytes are analyzed using a laser analytical flow cytometer confirming GLAST is greater than 90%. FACScan will be provided.
